Estrogen receptors in the rat uterus. Studies on the interaction of cytosol and nuclear binding sites.

چکیده
Studies on the interrelationship of binding sites for estrogens found in cytosol and nuclear fractions of the rat uterus show that 17/3-estradiol is f&t associated with a protein (9 S) present in the cytosol. The estrogen subsequently moves into the nucleus where it is bound to a protein (5 S) which can be extracted from the chromatin. Simultaneously with the movement of the estrogen into the nucleus, the binding protein of the cytosol disappears or loses its ability to bind estrogen. These and other experiments are discussed in terms of a hypothetical model in which estrogen binds to a cytosol receptor protein, bringing about a change in its conformation and its subsequent movement into the nucleus.

Estrogen receptors in ovary and uterus of immature hamster and rat: effects of estrogens.
Cytosolic and nuclear estrogen receptors in the ovary and uterus of immature rats and hamsters were determined to evaluate why exogenous estrogens were ineffective in stimulating follicular maturation in the hamster compared to the rat. Animals were injected sc with oil or single injection of 1 mg estradiol cyclopentylpropionate (ECP) on Day 23 or a daily injection of 2 mg diethylstilbestrol (D...
متن کاملAn endogenous inhibitor of [3H]estradiol binding to nuclear type II estrogen binding sites in normal and malignant tissues.
Previous studies from our laboratory have demonstrated that nonneoplastic tissues, such as rat uterus, contain an endogenous inhibitor of [3H]estradiol binding to nuclear type II sites. Since the stimulation of nuclear type II estrogen binding sites in the rat uterus is highly correlated with the stimulation of uterine growth and DNA synthesis, we have suggested that this inhibitor may be an en...
متن کاملUterine type II estrogen-binding sites are not of eosinophil origin.
A recent report by Lyttle et al. (Lyttle, C. R., Medlock, R. L., and Sheehan, D. M. (1984) J. Biol. Chem. 259, 2697-2700) suggested that nuclear type II sites in the rat uterus are of eosinophil origin and may represent [3H]estradiol binding to eosinophil peroxidase. To further evaluate this hypothesis we examined the response of nuclear type II sites to estrogen under conditions where eosinoph...
متن کاملNovel Small Molecules against Two Binding Sites of Wnt2 Protein as potential Drug Candidates for Colorectal Cancer: A Structure Based Virtual Screening Approach
Wnts are the major ligands responsible for activating Wnt signaling pathway through binding to Frizzled proteins (Fzd) as the receptors. Among these ligands, Wnt2 plays the main role in the tumorigenesis of several human cancers especially colorectal cancer (CRC). Therefore, it can be considered as a potential drug target. The aim of this study was to identify potential drug candidates ...
